On the completely opposite side of the current trend in posts.

Can a Mayor, during D&SS, choose a completely empty space of terrain, and simply build from the ground up? using EP? and of course his bonus parcels/population?

Nope, use of EP is for during Expo, not DSS. You would need to select an existing settlement to participate in DSS.

I should have worded it better, founding the town with your bonus parcels and population, then upgrading it later during expo.

As Stelaphina said, there's nothing to select in DSS if you build from scratch. But you can select a premade hamlet and build it up later in Expo for sure.
