Kings can have heirs, so they can rule over multiple lifetimes, yes. But they can also be overthrown, just as mayors can be voted out. Nothing is permanent in CoE and nothing lasts in perpetuity.
A noble who dies with a living child can specify that child as their heir and they can inherit the nobles titles.
HOWEVER before the player can take over their child they first have to have enough story points to buy into that NPC. If the player lacks sufficient SP the child will still hold the titles and such but the player will ha e to choose someone else to spark into.
It’s possible for a player to keep their titles through their heir, but it’s also possible for someone else to take over their title in various ways.
Nothing is permanent in this, but if a person/family puts in the effort they will be able to retain it for a long time, but they will have earned it.
Keeping in mind everything being said is theoretical, none of it has been put into practice so it remains to be seen how it all will work post launch.

I imagine it can also change within that 1 lifetime. I'm curious as to what circumstances could potentially strip someone of their title and lands. Can a king or queen punish a mayor or count etc by banishing them and confiscating what they own?
It also brings up my previous curiosity as to how punishments will work in Elyria.

It is fairly likely, though not certain, that kings will be kings for life -- at least character life. It is unlikely that many characters will survive the removal of their title.
But, seriously, this game has heirs and inheritances. A player can have their next character inherit. Can pick another player's character to inherit. May even be able to pick an NPC to inherit (we don't know).
They may even die without a clear heir -- which is bad when you own a shop, potentially disastrous (in a fun, war of succession kind of way) if a ruling royal does it.
